Description:
The research group “Molecular and Cellular Oncology” (Head: Prof. Stefan Fröhling) at the National Center for Tumor Diseases (NCT) Heidelberg and the German Cancer Research Center (DKFZ) is seeking an exceptionally talented and highly motivated PhD student to elucidate the biological function of rare cancer mutations. The project is part of the newly established research program “SyNC – Systematic analysis of mutations and Networks in Cancer”, which is a collaborative initiative between groups at Heidelberg University, DKFZ, NCT Heidelberg, and EMBL that aims at a comprehensive investigation of the function and relevance of uncharacterized genetic events and their intracellular networks in human tumors, with the ultimate goal to translate basic science discoveries into targeted cancer therapies.

The successful candidate will investigate the function of genetic alterations in human tumors and characterize associated phenotypic dependencies. Specifically, the PhD candidate will (i) establish cellular models of cancer-associated genetic alterations by genome engineering using CRISPR/Cas9 technology and/or lentiviral gene and shRNA transfer; (ii) extensively characterize these engineered cell lines phenotypically through, for example, assays for invasion, migration, anchorage independence, proliferation, and tumor growth in mice; (iii) profile these cell lines functionally using genome-wide shRNA and/or CRISPR/Cas9 libraries to identify co-dependencies and understand interactions within cellular networks; and (iv) mechanistically characterize the most interesting mutations and functional dependencies in detail in mouse models and primary patient samples.

With this project, the PhD candidate has the opportunity to work on the frontier of cancer genomics and to substantially enhance our understanding of cancer genotype-phenotype relationships and generate new starting points for exploiting specific tumor vulnerabilities through targeted drugs. In addition, the systematic investigation of cancer variants and associated functional dependencies will broaden our mechanistic understanding of many cellular processes.

Your profile:
· Applicants must hold a master’s degree in biology, biochemistry, biotechnology, or a related field
· The successful candidate is a highly motivated, ambitious, and hard-working individual with excellent grades who enjoys working in an international team and is passionate about making a difference through cancer research
· Strong communication and team working skills are required
· An excellent written and oral command of English is essential
· The ideal candidate has expertise in molecular and cellular biology, with specific focus on the techniques described above
